Analysis of arbuscular mycorrhizal (AM) fungal diversity through morphological characters of spores and intraradicular hyphae has suggested previously that preferential associations occur between plants and AM fungi. A field experiment was established to investigate whether AM fungal diversity is affected by different host plants in upland grasslands. Indigenous vegetation from plots in an unimproved pasture was replaced with monocultures of either Agrostis capillaris or Lolium perenne. Summary 1It is hypothesized that defoliation-induced changes in plant carbon allocation and root soluble C concentration modify rhizosphere C availability and, further, the abundance and activity of soil microbes and their grazers. To test this hypothesis, field-grown Lolium perenne swards were defoliated twice during their second growing season at two nitrogen availabilities (added N or no added N). Chemical and physical analysis, 27-d plant growth assays with carrot (Daucus carota) and Chinese cabbage (Brassica campestris var. chinensis), and 5-d phytotoxicity assays with Chinese cabbage and perennial ryegrass (Lolium perenne) were used to investigate the suitability of anaerobically digested poultry slaughterhouse waste for fertiliser in agriculture and the effect of aerobic post-treatment on the properties of the digested material. Rhizosphere soil was sampled in monocultures of Lolium perenne and Trifolium repens in June and October 2002, at two different nitrogen fertilisation levels (14 and 56 g N m−2 year−1) and under two pCO2 atmospheres (360 and 600 ppmv) at the Swiss FACE (Free Air Carbon dioxide Enrichment) site. Directly extracted soil DNA was analysed with restriction fragment length polymorphism (PCR-RFLP) by use of degenerated primers for the narG gene encoding the active site of the membrane-bound nitrate reductase. When soil nitrate levels are low, plants suffer nitrogen (N) deficiency but when the levels are excessive, soil nitrates can pollute surface and subsurface waters. Strategies to reduce the nitrate pollution are necessary to reach a sustainable use of resources such as soil, water and plant. Buffer strips and cover crops can contribute to the management of soil nitrates, but little is known of their effectiveness in semiarid vineyards plantations. Maģistra darba mērķis – izvērtēt vidi piesārņojošo metālisko elementu mobilitāti un biopieejamību barības ķēdes segmentā augsne-augs atkarībā no dažādu augsnes piedevu, piemēram, sapropelis, māls un arī uz sapropeļa un māla bāzes veidotu piedevu pievienošanas augsnēm. Sapropeļa kā augsnes piedevas ietekme uz augu attīstību tika vērtēta pēc dīgšanas un augu agrīnās attīstības testiem, izmantojot trīs kultūraugu (gurķu, tomātu, ganību airenes) sēklas. Darbā tika novērtēta Zn, Pb, Cd un Cu imobilizācija un biopieejamība, izmantojot augsnes ūdens un NH4NO3 ekstrakciju metodes.
